Is there a way to modify or update the contents of a Dropbox Transfer once it has been created?
Annoyingly, no, there isn't.
I've also discovered that if files hadn't fully uploaded before creating the transfer (i.e. you do it via Finder/Explorer while syncing) it wont sync everything.
