Create a new folder in Dropbox and use Selective Sync to remove that folder from your local computer. Add your 50GB+ file to your Dropbox folder and allow it to sync. Once the sync is complete, go on the Dropbox website and move the file into the folder you created and removed from your local computer. When you do that, the file will be removed from your computer but it will remain in the unsynced folder in your account online.

Are you trying to upload this file through our website? If so, I'm afraid that the upload limit for the site is 50 GB.
For larger files, we recommend using the Dropbox desktop app.
